About the Product
COMPOSITION
In 1 ml of the product contains:
- vitamin A (retinyl palmitate) — 20,000 IU
- vitamin D₃ (cholecalciferol) — 10,000 IU.
INDICATIONS FOR USE
·deficiency of vitamin A and D₃;
· supportive treatment of skin and mucous membrane diseases (including psoriasis, ichthyosis, and dermatoses resulting from progression of malignant processes).
DOSAGE AND ADMINISTRATION
Colefer is taken orally, dissolved in a spoonful of liquid.
Children aged 4 months to 12 months: 2 drops per day
Children aged 1 year to 18 years: 3 drops per day
Adults: 3 drops 2 times per day
Pregnant and lactating women: 3 drops 2 times per day. (No more than 4000 IU of vitamin A)

PHARMACOLOGICAL PROPERTIES
Pharmacodynamics
Vitamin A (retinol) is a fat-soluble vitamin essential for growth, epithelial differentiation, and mucosal integrity. It has antioxidant properties and neutralizes free radicals.
Retinol is involved in:
- formation of rhodopsin (night vision);
- metabolism of proteins, carbohydrates, and steroid hormones;
- regulation of thyroid function;
- stimulation of hepatic enzyme activity.
Vitamin D₃ and its metabolites maintain physiological calcium and phosphate levels in blood. Together with parathyroid hormone, they:
- enhance intestinal calcium absorption;
- regulate renal calcium excretion;
- modulate osteoclast activity.
This supports:
- bone mineralization;
- neuromuscular function.
Vitamins A and D₃ act synergistically via RXR and VDR receptors, regulating gene expression. This explains their use in dermatological conditions (psoriasis, T-cell lymphoma, keratoses).
Pharmacokinetics
Vitamin A:
- rapidly absorbed in the small intestine (60–90% bioavailability);
- stored in the liver as retinol esters;
- transported in blood bound to proteins;
- metabolized to retinal and retinoic acid;
- excreted via urine and bile.
Vitamin D₃:
- absorbed in the small intestine with bile acids (50–80% bioavailability);
- transported to the liver for hydroxylation;
- further metabolized in liver and kidneys;
- mainly excreted in bile, partly in urine.
CONTRAINDICATIONS
·hypersensitivity to retinol, cholecalciferol, or any excipients of the product;
·hypervitaminosis A and/or vitamin D₃;
·renal insufficiency;
·hypercalcemia;
·malabsorption syndrome;
·not recommended in infants under 4 months of age due to possible hypersensitivity reactions;
· concomitant use with other products containing vitamin A and/or vitamin D₃ is contraindicated.
ADVERSE REACTIONS
In patients with increased sensitivity to vitamin A and/or D₃ (rare) or during prolonged use of high doses, hypervitaminosis A and D₃ may develop.
The following may also occur:
- dry mouth
- increased calcium levels in blood and/or urine
- tissue calcification
- dizziness
- irritability
- angular cheilitis
- hair loss
- dry skin
- abdominal pain
- anemia
- subcutaneous edema
- bone and joint pain
These symptoms usually disappear completely after discontinuation of the vitamins.
Prolonged use of vitamin A may cause liver damage.
DRUG INTERACTIONS
Absorption of vitamin A may be reduced when used concomitantly with:
- cholestyramine, colestipol
- oral neomycin
- mineral oil
In such cases, an increased dose of vitamin A may be required.
Oral contraceptives increase plasma concentrations of vitamin A.
Concomitant use of vitamin A with other retinoids may increase its toxicity.
High doses of vitamin A (>50,000 IU) combined with tetracyclines may cause intracranial hypertension.
Concomitant use with alcohol may enhance hepatotoxic effects.
Long-term use of aluminum-containing antacids together with vitamin D₃ may increase blood aluminum levels, leading to toxic effects on bone tissue.
SPECIAL PRECAUTIONS
- Overdose should be avoided.
- During treatment, foods rich in vitamin A (e.g., liver) should not be consumed.
- Use with special caution in pregnant women.
- Caution is required in patients with:
- immobilization
- use of thiazide and other diuretics
- urolithiasis
- heart diseases
- use of cardiac glycosides
- In infants with congenitally small anterior fontanelle, Colefer should be used with particular caution.
Pregnancy and lactation
High doses of vitamins A and D₃ should be avoided due to potential fetal toxicity.
During pregnancy and breastfeeding, the daily dose of vitamin A must not exceed 4000 IU.
