COMPOSITION

1 capsule contains:

• • Alverine citrate — 60 mg 
  • Simethicone — 300 mg. 

INDICATIONS FOR USE 

• • symptomatic treatment of functional intestinal disorders accompanied by abdominal bloating and flatulence (in particular, irritable bowel syndrome); 
  • preparation for radiological, ultrasound, or instrumental examination of the abdominal organs. 

DOSAGE AND ADMINISTRATION

Capsules are taken orally whole, without chewing, with sufficient water.

The drug is taken at a dose of 1 capsule 2–3 times daily before meals for 3–4 weeks.

PHARMACOLOGICAL PROPERTIES

Pharmacodynamics

Simespasmil is a combination drug whose effects are determined by its components. It has antispasmodic activity and reduces intestinal gas formation.

Alverine is an antispasmodic agent that acts directly on the smooth muscles of the gastrointestinal tract. Its mechanism involves inhibition of phosphodiesterase and accumulation of intracellular cyclic adenosine monophosphate, followed by blockade of calcium channels in smooth muscle cells.

Simethicone reduces the surface tension of gas bubbles, causing them to break down. The released gases are partly absorbed in the intestine and eliminated through intestinal peristalsis.

Pharmacokinetics

Alverine citrate is very slowly absorbed from the gastrointestinal tract and has low bioavailability after oral administration. Maximum plasma concentration corresponds to about 0.15% of the administered dose. Plasma levels remain stable for 6 hours.

Simethicone, due to its physiological and chemical inertness, is not absorbed in the body and is excreted unchanged via the gastrointestinal tract.

After oral administration, the effect develops within 30–60 minutes and lasts for 3–4 hours.

CONTRAINDICATIONS

• • hypersensitivity to any component of the drug; 
  • intestinal obstruction; 
  • pregnancy and lactation; 
  • children and adolescents under 18 years of age.

• • rash; 
  • itching; 
  • swelling of the face, tongue, and larynx; 
  • breathing disorders; 
  • allergic reactions; 
  • headache; 
  • dizziness; 
  • nausea; 
  • constipation.

DRUG INTERACTIONS

No cases of drug interactions of Simespasmil with other medicinal products have been reported.

SPECIAL PRECAUTIONS

Caution should be exercised if signs suggestive of intestinal obstruction appear.
