COMPOSITION

1 capsule contains:

ursodeoxycholic acid — 300 mg

INDICATIONS FOR USE 

·uncomplicated gallstone disease; 

·non-alcoholic steatohepatitis; 

·acute and chronic viral hepatitis; 

·toxic liver damage; 

·chronic active hepatitis; 

·alcoholic liver disease; 

·primary biliary cirrhosis; 

·primary sclerosing cholangitis; 

·cystic fibrosis; 

·congenital intrahepatic biliary atresia; 

·  cholestasis during parenteral nutrition; 

·biliary reflux gastritis and reflux esophagitis; 

·biliary dyspeptic syndrome; 

·biliary tract dyskinesia; 

·prophylaxis: 

    • liver damage due to hormonal contraceptives or cytostatic therapy; 
    • formation of gallstones in obese patients during rapid weight loss

DOSAGE AND ADMINISTRATION

    • For acute and chronic liver diseases and gallstone dissolution: 
    • dose: 10–15 mg/kg body weight
    • treatment duration: from several months up to 2 years
    • For biliary reflux gastritis and reflux esophagitis: 
    • dose: 300 mg
    • treatment duration: 10–14 days.

PHARMACOLOGICAL PROPERTIES

Choludexan – hepatoprotector.

Pharmacological effects:

    • cholelitholytic; 
    • choleretic; 
    • hepatoprotective; 
    • hypocholesterolemic; 
    • immunomodulatory. 

Pharmacokinetics:

    • absorbed in the intestine; 
    • Cmax = 5.5 mmol/L reached after 1 hour
    • metabolized in the liver into taurine and glycine conjugates, excreted in bile; 
    • plasma protein binding up to 99%
    • crosses the placental barrier; 
    • 50–70% excreted via intestines.

CONTRAINDICATIONS

·hypersensitivity to the drug; 

·acute inflammatory diseases of the gallbladder and bile ducts; 

·  non-functioning gallbladder.

ADVERSE REACTIONS

·diarrhea; 

·  back pain.

DRUG INTERACTIONS

·ntacids containing aluminum ions and ion-exchange resins (e.g., cholestyramine) reduce the absorption of the drug. 

·  Lipid-lowering agents (especially clofibrate), estrogens, neomycin, and progestins may reduce the ability of the drug to dissolve cholesterol gallstones.

SPECIAL PRECAUTIONS

For successful dissolution of gallstones, they should:

    • consist purely of cholesterol; 
    • be radiolucent (do not cast a shadow on X-ray); 
    • have a size of no more than 15–20 mm.
