INDICATIONS FOR USE 

-Arterial hypertension 

-Ischemic heart disease (stable angina, Prinzmetal’s angina) 

-Chronic heart failure (as part of combination therapy) 

-Diabetic nephropathy  

DOSAGE AND ADMINISTRATION

Amlipin should be taken orally, 1 tablet once daily. If necessary, the daily dose may be increased to 2 tablets. The medication can be taken with or after meals.

PHARMACOLOGICAL PROPERTIES

Amlipin is a combined antihypertensive medication whose pharmacological effect is based on the combination of: Amlodipine (a selective calcium channel blocker) and Lisinopril (an ACE inhibitor). After oral administration, amlodipine is slowly and almost completely (about 90%) absorbed from the gastrointestinal tract. Maximum concentration (Cmax) is reached within 6–10 hours. The half-life (T½) is 35–50 hours. 

Lisinopril is well absorbed from the gastrointestinal tract regardless of food intake. Its bioavailability is 25–50%. Maximum concentration (Cmax) is reached within 6 hours. 

The half-life (T½) is approximately 12 hours. 

The prolonged circulation of both active substances allows once-daily dosing.

CONTRAINDICATIONS

Amlipin is contraindicated in patients with known hypersensitivity to any of its components. Сhildren under 18 years of age

ADVERSE REACTIONS

Possible side effects include dizziness, headache, nausea, fatigue, edema, and allergic reactions. Symptomatic hypotension may occur in very rare cases.

DRUG INTERACTIONS

When used concomitantly with other antihypertensive agents, an enhanced hypotensive effect may occur. Nonsteroidal anti-inflammatory drugs (NSAIDs) may reduce the antihypertensive effects of amlodipine and lisinopril. Co-administration of amlodipine with sympathomimetics may decrease its antihypertensive efficacy, while use with lithium preparations may increase the risk of neurotoxicity.

 Concurrent use of lisinopril with potassium-sparing diuretics may lead to hyperkalemia; with lithium preparations, it may reduce lithium elimination from the body.

SPECIAL PRECAUTIONS

Use with caution in patients with impaired liver function, aortic stenosis, or chronic heart failure. It should not be used in patients with acute myocardial infarction in cases where there is a risk of severe hemodynamic disturbances associated with vasodilator therapy, or in patients with impaired renal function. The safety of combined use of amlodipine and lisinopril during pregnancy and lactation has not been established.
