COMPOSITION

1 capsule contains:

Mebeverine hydrochloride 200 mg. 

INDICATIONS FOR USE 

In adults:

    • irritable bowel syndrome (colon); 
    • gastrointestinal spasms, including those associated with organic disease; 
    • intestinal colic; 
    • biliary colic. 

In children over 12 years of age:

    • functional gastrointestinal disorders associated with abdominal pain. 

DOSAGE AND ADMINISTRATION

Duskonal Retard is taken orally, swallowing the capsule whole, 20 minutes before meals, with sufficient liquid.

Adults (including elderly patients) are prescribed 200 mg twice daily.

Duration of treatment is not limited. If a dose is missed, treatment should be continued as prescribed; the missed dose should not be taken in addition to the next scheduled dose.

No dose adjustment is required in elderly patients or in patients with renal and/or hepatic impairment.

PHARMACOLOGICAL PROPERTIES

Duskonal Retard is a myotropic antispasmodic. Mebeverine directly relaxes smooth muscle, mainly in the gastrointestinal tract, relieving spasm without significantly affecting normal intestinal motility.

Its mechanism involves blockade of sodium channels in the cell membrane, reducing Na⁺ influx, inhibition of Ca²⁺ entry through slow channels, and reduction of K⁺ efflux, resulting in decreased muscle contractility. It has no anticholinergic effects.

After oral administration, mebeverine is rapidly and completely absorbed. The prolonged-release formulation allows twice-daily dosing. No significant accumulation occurs with repeated dosing.

Mebeverine is extensively metabolized by esterases to veratric acid and mebeverine alcohol. The main circulating metabolite is demethylated carboxylic acid (DMCA). The elimination half-life of DMCA at steady state is 5.77 hours.

Bioavailability is approximately 97%. The parent compound is not excreted unchanged; metabolites are eliminated mainly via the kidneys.

CONTRAINDICATIONS

·hypersensitivity to the components of the drug; 

·  children under 12 years of age.

ADVERSE REACTIONS

Dizziness (in some cases), diarrhea or constipation.

Allergic reactions: angioedema (Quincke’s edema), urticaria, exanthema, and facial swelling.

DRUG INTERACTIONS

There are insufficient data on the use of mebeverine in pregnant women. No teratogenic effects were observed in animal studies. It is unknown whether mebeverine or its metabolites are excreted in breast milk. Therefore, breastfeeding is not recommended during treatment with this medicine.

SPECIAL PRECAUTIONS

No interactions of mebeverine with other medicinal products have been identified.
